Illumina Demonstrated 16S Protocol describes a method for preparing samples for sequencing the variable V3 and V4 regions of the 16S rRNA gene. This protocol can also be used for sequencing other regions with different region‐specific primers. This protocol combined with a bench top sequencing system, on‐board primary analysis, and secondary analysis using MiSeq Reporter or BaseSpace, provides a comprehensive workflow for 16S rRNA amplicon sequencing.
