ProP-PD/PDZ-Seq
Proteomic peptide-phage display (ProP-PD) is a protocol that identifies short linear motif (SLiM) interactions or PDZ domains (PDZ-Seq ). In this method, C-terminal sequences are first identified, an oligo library is created, followed by construction of a phage display library, which is hybridized to immobilized bait proteins. Sequences of display phages that interact with the bait protein are isolated, sequenced and counted. Deep sequencing provides detailed information pertaining to the motifs that interact with specific proteins.
